THP-1 nuclear extract (TPA stimulated)


Active Motif's THP-1 (TPA stimulated) nuclear extract was prepared from a cell culture of the human acute monocytic leukemia THP-1 cell line. The THP-1 cell line was originally derived from the peripheral blood of a 1 year old body with acute monocytic leukemia. THP-1 cells share similar properties to human monocyte-derived macrophages. These cells differentiate into mature macrophages (phagocytes) when grown in the presence of DMSO or phorbol 12-myristate 13-acetate (TPA, also known as PMA). When stimulated with DMSO or vitamin D3, THP-1 cells adhere to the walls of cell culture flasks, and are therefore frequently utilized as in vitro model systems to study foam cell formation in relation to atherosclerosis. The THP-1 cell line is characterized by the expression of Fc and C3b receptors, the production of IL-1, and the lack expression of cytoplasmic and surface immunoglobulins. In addition, these cells are phagocytic, produce lysosymes and stain positive for alpha-napthyl butyrate esterase. Because of these properties, they are often used in research to study monocyte differentiation.
