Full-length recombinant human CK1 epsilon was expressed by baculovirus in Sf9 insect cells using an N-terminal GST tag. CK1 epsilon is a member of the CK1 family of serine/threonine protein kinases, which play an important role in diverse cell processes, including DNA replication and repair. CK1 epsilon is a regulator of Yes-associated protein (YAP) transcription coactivator, which is a key regulator of organ size and a candidate human oncogene. CK1 epsilon is activated by CCK2R, and this then phosphorylates PKD2 at Ser244. Phosphorylation of PKD2 leads to its nuclear accumulation and efficient phosphorylation of nuclear PKD2 substrates in human gastric cancer cells (1). CKI epsilon can phosphorylate topoisomerase (topo) IIalpha at serine-1106, and this regulates the enzyme activity and sensitivity to topo II-targeted drugs (2). ADP-Glo Kinase Assay is a luminescent kinase assay that measures ADP formed from a kinase reaction; ADP is converted into ATP, which is a substrate in a reaction catalyzed by Ultra-Glo Luciferase that produces light. The luminescent signal positively correlates with ADP amount and kinase activity. The assay is well suited for measuring the effects chemical compounds have on the activity of a broad range of purified kinases, making it ideal for both primary screening as well as kinase selectivity profiling. The ADP-Glo Kinase Assay can be used to monitor the activity of virtually any ADP-generating enzyme (e.g., kinase or ATPase) using up to 1mM ATP. Kinase Enzyme System contains: Kinase: CK1 epsilon, 10ug (Human, recombinant full-length). MW: ~72kDa. Substrate: Casein, dephosphorylated; native protein purified from bovine milk. Other: Reaction Buffer, DTT. CK1 epsilon NCBI Database Entry: www.ncbi.nlm.nih.gov/gene/1454/. Visit www.promega.com/kinase/ to see all Kinase Enzyme Systems.
