Recombinant human FLT1 (amino acids 784-end) was expressed by baculovirus in Sf9 insect cells using an N-terminal GST tag. FLT1, also known as fms-like tyrosine kinase 1, is a member of the VEGFR family and is related to the oncogene ROS. The tyrosine kinase activity of FLT1 has been shown to play a key role in control of cell proliferation and differentiation. FLT1 is a receptor for vascular endothelial growth factor (VEGF), and it has been observed that the ratio of FLT1 mRNA to VEGF mRNA correlates with tumor angiogenesis and prognosis in non-small cell lung cancers. In addition to the full-length receptor, FLT1 gene also encodes a soluble form of receptor due to alternative splicing. Both the full length and soluble form of FLT1 show strong binding affinity for VEGF. ADP-Glo Kinase Assay is a luminescent kinase assay that measures ADP formed from a kinase reaction; ADP is converted into ATP, which is a substrate in a reaction catalyzed by Ultra-Glo Luciferase that produces light. The luminescent signal positively correlates with ADP amount and kinase activity. The assay is well suited for measuring the effects chemical compounds have on the activity of a broad range of purified kinases, making it ideal for both primary screening as well as kinase selectivity profiling. The ADP-Glo Kinase Assay can be used to monitor the activity of virtually any ADP-generating enzyme (e.g., kinase or ATPase) using up to 1mM ATP.
