IGF1R Kinase Enzyme System
Recombinant human IGF1R (amino acids 960-end) was expressed by baculovirus in Sf9 insect cells using an N-terminal His tag. IGF1R (Insulin-like Growth Factor 1) is a transmembrane tyrosine kinase receptor that is activated by IGF-1 and by the related growth factor IGF-2. IGF1R mediates the effects of IGF-1 and plays an important role in growth and anabolic effects in adults. The IGF1R is implicated in several cancers, most notably breast cancer, where it is highly overexpressed and functions as an anti-apoptotic agent by enhancing cell survival. In many instances, the anti-apoptotic properties of IGF1R overexpression allow cancerous cells to resist the cytotoxic properties of chemotherapeutic drugs or radiotherapy. ADP-Glo Kinase Assay is a luminescent kinase assay that measures ADP formed from a kinase reaction; ADP is converted into ATP, which is a substrate in a reaction catalyzed by Ultra-Glo Luciferase that produces light. The luminescent signal positively correlates with ADP amount and kinase activity. The assay is well suited for measuring the effects chemical compounds have on the activity of a broad range of purified kinases, making it ideal for both primary screening as well as kinase selectivity profiling. The ADP-Glo Kinase Assay can be used to monitor the activity of virtually any ADP-generating enzyme (e.g., kinase or ATPase) using up to 1mM ATP.
