MYO3 beta Kinase Enzyme System
Recombinant human MYO3 beta (1-326) was expressed by baculovirus in Sf9 insect cells using an N-terminal GST tag. MYO3 beta is a member of the Class III myosins that are actin-dependent motor proteins containing an amino-terminal kinase domain. MYO3 beta contains an N-terminal kinase domain, followed by motor, neck and tail domains. The MYO3 beta gene generates a variety of splice variants that contain 1 or 2 calmodulin-binding (IQ) motifs in the neck domain and 1 of 3 domains in the tail domain. Northern blot analysis shows expression of a 7kb MYO3 beta transcript in the human retina but not in the RPE cell line (1). The MYO3 beta gene transcript is also detected in the kidney, intestine and testis. The MYO3 beta gene maps to chromosome 2q31.1-q31.2 by genomic sequence alignment. ADP-Glo Kinase Assay is a luminescent kinase assay that measures ADP formed from a kinase reaction; ADP is converted into ATP, which is a substrate in a reaction catalyzed by Ultra-Glo Luciferase that produces light. The luminescent signal positively correlates with ADP amount and kinase activity. The assay is well suited for measuring the effects chemical compounds have on the activity of a broad range of purified kinases, making it ideal for both primary screening as well as kinase selectivity profiling. The ADP-Glo Kinase Assay can be used to monitor the activity of virtually any ADP-generating enzyme (e.g., kinase or ATPase) using up to 1mM ATP. Kinase Enzyme System contains: Kinase: MYO3 beta, 10ug (Human, recombinant; amino acids 1-326). MW: ~63kDa. Substrate: Native Swine Myelin Basic Protein (MBP). Other: Reaction Buffer, DTT.
